Adeno-associated virus (AAV) packaging services refer to customized development and manufacturing activities in which a customer-provided or jointly designed ITR-containing construct is packaged into a recombinant AAV vector carrying a gene of interest, gene-silencing sequence, gene-editing component or reporter gene. The service workflow generally includes vector design and construction, plasmid preparation, cell culture and transfection, vector harvest, purification, concentration, aliquoting and quality testing, covering research-grade, preclinical-grade and GMP-grade projects. Key upstream inputs include transfer plasmids, Rep/Cap plasmids, helper plasmids, HEK293 or Sf9 cells, culture media, transfection reagents, nucleases, chromatography resins, filtration consumables and analytical reagents. Major downstream customers include universities, research institutes, hospital research platforms, gene therapy developers, biotechnology companies, pharmaceutical companies and CROs, with applications spanning basic research, animal studies, preclinical evaluation, IND submissions, clinical development and commercial manufacturing. The industry's overall gross margin is generally estimated at approximately 32%-48%.
The global market consists of research-grade packaging providers, specialized AAV CROs, integrated cell and gene therapy CDMOs and large biopharmaceutical service groups operating dedicated AAV platforms. Research-grade projects are numerous, relatively short and fragmented, with competition centered on serotype coverage, titer, purity, turnaround time and price. Preclinical and GMP programs are less frequent but require considerably greater investment in process scale-up, analytical methods, batch consistency, quality systems and regulatory documentation. Providers capable of maintaining process continuity from vector design and small-scale packaging through GMP and commercial manufacturing generally develop stronger customer retention, particularly after a program enters clinical development.
Demand is supported by gene-function research, rare-disease and neurological gene therapy, engineered-capsid screening, gene-editing delivery and greater outsourcing by biotechnology companies. Different AAV capsids enable gene delivery to the eye, liver, central nervous system, muscle and other tissues, while dual- and multi-vector systems extend the technology to larger or more complex payloads. The FDA's 2026 approval of the first dual-AAV gene therapy for inherited hearing loss demonstrates the continued expansion of AAV into new vector configurations and therapeutic areas, supporting demand for dual-vector development, analytics and scalable manufacturing.
Adherent HEK293 triple-plasmid transfection remains widely used for research and smaller programs, while suspension HEK293 systems have become the principal route for clinical and commercial scale-up. Baculovirus-Sf9 systems are used in selected large-scale processes. Stable producer-cell lines, dual-plasmid systems and plasmid-reduced technologies are being developed to lower raw-material costs and improve consistency. Downstream competition is moving beyond vector-genome titer toward full-capsid enrichment, genome-integrity control and tighter reduction of host-cell proteins, residual DNA, residual plasmids and replication-competent AAV.
The principal constraints include gene therapy financing and pipeline reprioritization, high clinical attrition, underutilized GMP facilities, elevated manufacturing costs and the immunological and safety limitations of AAV. Serious liver-related safety events led the FDA to restrict the use of a major AAV gene therapy in 2025, increasing the focus on dose selection, impurity control, clinical monitoring and long-term follow-up. At the same time, the FDA introduced a more flexible approach to certain cell and gene therapy CMC requirements in 2026, which may help selected rare-disease programs progress more efficiently but does not reduce expectations for identity, potency, purity and manufacturing consistency.
