Adeno-associated virus (AAV) is a type of parvovirus that has the ability to infect both dividing and non-dividing cells. It usually relies on co-infection with adenovirus or herpes virus to help it replicate and amplify in vivo. Recombinant adeno-associated virus (rAAV) vectors only need to carry the cis-element ITR to produce recombinant AAV particles. Due to the lack of Rep gene, the recombinant AAV viral genome does not integrate into the host genome and exists as an extrachromosomal free body for a long time. These characteristics make AAV viral vectors a gene delivery tool with great application prospects. Compared with other viral vectors, AAV has many advantages: 1) Strong diffusibility: rAAV is small in size and high in titer. Compared with adenovirus and lentivirus, it has higher diffusivity and can cross the blood-brain barrier, making it an ideal tool for infecting the nervous system; 2) High specificity: rAAV has multiple serotypes, and different serotypes have different recognition and infection abilities for different tissues. It is a tool virus preferred by animal experiments; 3) High safety: rAAV persists outside the chromosome, reducing the risk of mutation, and has low immunogenicity, so it can be directly used for human gene therapy.
