Triacetyl ribose is an important intermediate for the synthesis of capecitabine, and the capecitabine bulk drug can be prepared in three steps through a route reported in the literature. According to reports in the literature, the synthesis of triacetyl ribose is mainly based on the following two methods: one is using 1-methyl-5-deoxyribose as an intermediate, and triacetyl ribose is obtained through a two-step reaction; the second is using 5-deoxy-D -Ribose is an intermediate and triacetyl ribose is obtained by one-step acetylation. According to the synthetic method reported above, the prepared triacetyl ribose is a mixture of α and β isomers. The β isomer participates in the next reaction to prepare capecitabine. The α isomer is the impurity produced, and the α is different. The ratio of the structure is 3~30%, and the ratio of the α isomer will not be higher. The pure β isomer can be obtained by column chromatography or recrystallization at this isomer ratio.
