Endopeptidases are hydrolases that cleave peptide bonds within the protein or polypeptide chain, unlike exopeptidases which cleave from the ends. Their cleavage products are small polypeptide fragments (arginine, peptone), rather than free amino acids. These enzymes exhibit high substrate specificity; a single protease typically recognizes and cleaves peptide bonds formed by specific amino acid residues, such as trypsin specifically cleaving the carboxyl-terminal peptide bond of arginine (Arg) or lysine (Lys).
Raw material costs account for approximately 30%-40% of the total production cost of endopeptidases. Core raw materials include microbial strains (production strains or recombinant plasmid hosts) and fermentation media (carbon sources such as glucose/corn starch, nitrogen sources such as soybean flour/yeast extract/peptone, inorganic salts, and metal ions). Carbon and nitrogen sources, as bulk raw materials, account for a major portion of raw material costs. Using agricultural byproducts such as corn leaching solution (CSL) and wheat bran can significantly reduce costs. For recombinant expression products, high-purity enzyme preparations also require expensive chromatographic media (ion exchange, hydrophobic chromatography) and lyophilization protectants; these downstream purification costs can account for more than 50% of the total cost. In addition, energy consumption (temperature control, stirring, sterilization), equipment depreciation (fermenters, ultrafiltration systems), and quality control (activity assays, stability tests) during production are also significant cost components. Large-scale production (ton-level) can reduce unit costs to tens of yuan per kilogram by amortizing fixed costs, while small-batch high-purity research-grade products (such as sequencing-grade Lys-C) require multiple chromatographic purifications and strict quality control, resulting in costs as high as several thousand yuan per milligram.
