Long Read Sequencing (LRS) represents a paradigm shift in genomics, moving from reconstructing the genome like a puzzle to reading it in near-contiguous pieces. Unlike Short Read Sequencing (SRS), which fragments DNA into pieces 150-300 base pairs long (requiring complex computational assembly), LRS reads single molecules in lengths of 10,000 to over 1,000,000 base pairs.

1. Structural Variant (SV) Resolution (Clinical Driver)

Context: Short reads detect SNPs (single nucleotide variants) well. They miss 50-70% of structural variants (deletions, insertions, inversions, duplications >50bp).

Clinical Implication: SVs are implicated in autism, schizophrenia, epilepsy, cancer, and developmental disorders.

Driver: LRS detects SVs directly. No inference. The variant is seen in the read alignment.

2. Phasing & Haplotype Resolution (Mendelian Genetics)

The Problem: Humans are diploid. Short reads mix maternal and paternal alleles.
