Laser Microdissection, also known as LMD or LCM (Laser Capture Microdissection), is a contact- and contamination-free method for isolating specific single cells or entire areas of tissue from a wide variety of tissue samples. Laser microdissection is now used in a large number of research fields, e.g. neurology, cancer research, plant analysis, forensics or climate research. The method is meanwhile also applied for manipulation of cell cultures or for microengraving of coverslips. Laser Microdissection System are perfect tools to optimize DNA-Workflows (Genomics), RNA-Workflows (Transcriptomics) and Proteomic-Workflows as they allow to precisely define and collect pure starting material for analysis under visual control.
Laser microdissection offers a precise and contamination-free solution for the isolation and selection of single cells or tissue. The tissue samples can be embedded, sectioned, and stained according to conventional methods of preparation. Paraffin sections, frozen sections, smear preparations, chromosome specimens, and cell cultures are all suitable for laser microdissection. The area selected for dissection is drawn on the PC screen and automatically separated from the surrounding tissue with a laser beam. Fluorescence-labelled specimens can also be dissected using special filter cubes which transmit the full spectrum of laser light. The dissectate is then immediately transported to a collection device (in different ways, depending on the manufacturer of the system) for further examination. The precision of the laser-cutting process is optically coupled to the chosen magnification. A higher magnification automatically results in a finer step width, as the laser beam and its movement are reduced by the same degree as the field of view. No other work steps are required in this case. Not only single cells, but also larger areas can be excised in a single pass. When transferring the dissectate to a collection device, there is no risk of contact or contamination. Laser microdissection (LM) has become widely democratized over the last fifteen years. Instruments have evolved to offer more powerful and efficient lasers as well as new options for sample collection and preparation. Technological evolutions have also focused on the post-microdissection analysis capabilities, opening up investigations in all disciplines of experimental and clinical biology, thanks to the advent of new high-throughput methods of genome analysis, including RNAseq and proteomics, now globally known as microgenomics, i.e. analysis of biomolecules at the cell level. In spite of the advances these rapidly developing methods have allowed, the workflow for sampling and collection by LM remains a critical step in insuring sample integrity in terms of histology (accurate cell identification) and biochemistry (reliable analyzes of biomolecules).
