A cDNA library is a combination of cloned cDNA fragments inserted into a collection of host cells. It uses mRNA as a template, catalyzed by reverse transcriptase, reverse transcribed into cDNA in vitro, and ligated with an appropriate vector (commonly used phage or plasmid vector) to transform the recipient bacteria. Each bacteria contains a piece of cDNA and can reproduce and expand. In this way, the collection of cDNA clones containing all the mRNA information of the cell is called the cDNA library of the tissue cell. cDNA libraries specifically reflect the genes encoding proteins expressed in specific tissues or cells at specific developmental stages, so cDNA libraries are tissue- or cell-specific.

A cDNA library, also known as a complementary DNA library or expression library, is a collection of DNA fragments that represent the entire set of genes expressed by an organism, tissue, or cell at a specific point in time. These libraries are essential tools in molecular biology and genomics research, allowing scientists to study gene expression patterns and isolate specific genes of interest. The use of NGS technologies for high-throughput sequencing of cDNA libraries has increased, enabling researchers to analyze gene expression at a comprehensive level. Single-cell RNA sequencing (scRNA-seq) techniques have gained prominence, leading to the development of specialized cDNA libraries for studying gene expression at the single-cell level.
