Molecular hybridization is a technique for determining the sequence of single stranded nucleic acid bases. The basic principle is to form detectable double helix fragments between the tested single stranded nucleic acid and a known sequence of single stranded nucleic acid (called a probe) through base pairing. Currently, solid-phase hybridization is widely used. This method involves first binding the single stranded nucleic acid sample to the membrane (if it is double stranded, it must be denatured into a single stranded), and then hybridizing it with the labeled probe in the solution. Hybrid buffer usually refers to a solution commonly used in molecular biology hybridization experiments. It is usually used to maintain a specific pH value and contains some specific salts to maintain the stability and activity of biological molecules (such as DNA, RNA, or proteins) during the experimental process.
